JEFFREY'S MACERATION METHOD
PROCEDURE  TIME
PREPARATION
 STAINING MACERATED MATERIAL
1.  Cut material into small slices ca. 300 um thick.

2.  Place material in Maceration Fluid under vacuum to ensure all air is removed.

3. Use a glass rod to gently crush the material. 

4.  Centrifuge material and replace Maceration Fluid with water

2. Various times but cells should start separating by ca. 24 hrs.  Woody material should be placed in 40 degree oven.

3.  If material does not crumble easily, replace Maceration Fluid with fresh stock.

4. Repeat at least 3 times to insure material is acid free

 Equal parts of

1.  10% Aqueous Nitric Acid

2.  10% Aqueous Chromic Acid
 

 

 1.  Use a Pasteur Pipet to transfer a portion of the cell suspension to a centrifuge tube.

2.  Stain with your desired dye.

3.  Centrifuge.

4.  Remove dye solution.

5.  Wash in appropriate solvent. 

6.  Centrifuge to concentrate cells. 

7.  Mount on slide.
