Black Expanding Blue (acid hydrolyzes cellulose to hydrocellulose which reacts with IKI) |
1. Place sections in drop of IKI
solution on slide and cover with coverslip.
2. Allow drop of 65% Sulphuric Acid to diffuse underneath one side of coverslip while watching for swelling of cellulose. |
1. IKI solution = 1 g I and 3 g
KI in 300 ml of dH2O.
2. 65% aqueous Sulphuric Acid |
